CF065 is recombinant human PDGF-AA expressed in E. coli, filtered through 0.22μ membrane filter and lyophilized. PDGFs act as potent mitogens for a variety of cell types including smooth muscle cells. Platelet- Derived Growth Factor (PDGF) family of heparin binding growth factors consists of 5 members - PDGF-AA, PDGF-BB, PDGF-AB, PDGF-CC and PDGF-DD. These are disulfide-linked dimers consisting of two 12.0 - 13.5kDa polypeptide chains, designated PDGF-A, PDGF-B, PDGF-C and PDGF-D chains. PDGFR-α and PDGFR-β are the two signaling receptors for PDGFs. PDGFR-α is a high affinity receptors for all forms of PDGF whereas PDGFR-β interacts only with PDGF-BB, PDGF-AB and PDGF-DD. PDGF-AA is a homodimer of made up of 2 chains of polypeptide PDGF-A. Binding of PDGF-AA to its receptor on cells elicits a variety of cellular responses such as hyperplasia, fibroblast migration and embryonic development. PDGFs are strongly chemotactic for human monocytes and neutrophils. At higher concentration, they act as chemoattractant for fibroblasts and smooth muscle cells.
