SPS Agar is recommended for detection of Clostridium perfringens in food samples. This medium has tryptone and yeast extract that supply nitrogenous compounds, vitamin B complex and other essential growth nutrients. Polymyxin B and sulphadiazine inhibit a wide variety of gram-positive and gram-negative bacteria. Organisms reduces sulphite to sulphide which reacts with iron of ferric citrate to form a black precipitate of iron sulphide and hence the colonies appear black. Clostridium perfringens and Clostridium sporogenes shows black colored colony on this medium.
